Description
ISRIB (Integrated Stress Response Inhibitor) is a potent cognitive enhancer first discovered in 2013 by Carmela Sidrauski in the laboratory of Peter Walter at UCSF. It was identified through a high-throughput screen for compounds that could maintain protein synthesis even under cellular stress.
How It Works
ISRIB acts as a “molecular staple.” It binds to and stabilizes the eIF2B complex, which is a critical regulator of protein synthesis. By stabilizing this complex, ISRIB reverses the inhibitory effects of eIF2α phosphorylation, effectively “turning back on” the cell’s machinery for making new proteins—a process usually shut down during the Integrated Stress Response (ISR).
